Should be no problem. It's less practical though I think because you
have to deal with an extra cable that could come loose. And I think the
chance of running into interference issues could become bigger.
I am currently using a Pisound on a Raspberry Pi 4,
and it is working great. But it does use up all the gpio pins and I do not need its MIDI
facilities. I would like to be able to use other SBCs, also.

things improved?
Yes. Not only on the USB side of things but a Pi 4 is also way more
performant then a Pi 1. And already a decade ago latency was acceptable
with a Pi 1 and a USB interface, I was pioneering around 2013 with a Pi
1 and an Edirol UA-25 with 8ms system latency. Pi 1 was running Guitarix
and I had a Boss FC-50 hooked up to switch between effects (echo,
chorus, distortion). Worked pretty well actually.
