Don't see how. You need to do two
measurements anyway, no matter
how it's done, then add or substract.
Thinking again, you only need one measurement:
jack_delay -> pa_source -> audacity -> pa_sink -> system:playback ->
analog cable -> system_capture -> jack_delay
